马来酸桂哌齐特的改进合成

Abstract: 以(E)3,4,5-三甲氧基肉桂酸为原料,在-10 ℃经活化试剂氯甲酸乙酯活化后与1-[(1-四氢吡咯羰基)甲基]哌嗪(Ⅲ)"一锅煮"法合成得到桂哌齐特游离碱(Ⅳ),再与马来酸成盐后制得马来酸桂哌齐特(Ⅰ),以四氢吡咯计总收率为50.7%.中间体1-[(1-四氢吡咯羰基)甲基]哌嗪(Ⅲ)也从四氢吡咯经两步反应得到,利用化合物Ⅲ和哌嗪在丙酮中的溶解度差异进行后处理操作,产物Ⅲ质量分数可达到98%以上.
