SULFATED PACHYMARAN PROTECTS PC12 CELLS AGAINST MPP+-INDUCED INJURY

摘要:PC12 cells treated with 0.5 mM/MPP+ for 24 h were used as PD model in vitro to evaluate the neuroprotective effects of sulfated pachymaran (SP). Cell viability and apoptosis were detected by MTT assay, inverted microscope and Hoechst 33342 staining, respectively. Expression levels of Bcl-2, Bax and caspase-3 were detected by Western blotting. SP at the concentrations from 0.5 to 4 mg/mL is safe to PC12 cells. MPP+ treatment significantly decreased the cell viability, the expressions of Bcl-2 and Caspase-3 (P<0.01) and increased the expression of Bax (P<0.05). The pretreatment of 2 mg/mL SP significantly contributed to the restoration of cell viability (P<0.05), the up regulation of Bcl-2 (P<0.01) and Caspase-3 (P<0.01), and it also inhibited the overexpression of Bax (P<0.05). SP may inhibit apoptosis of MPP+-induced PC12 cells through the mitochondria-dependent Caspase pathway.
