Abstract:The whole cells of naphthalene-degrading strain Comamonas sp.MQ were used for the transformation of indole to indigo.The results showed that strain MQ possessed excellent activities for indigo production,and the maximum yield of indigo could be obtained within 4 h.Based on the analysis of thin-layer chromatography(TLC) and high performance liquid chromatography(HPLC),the blue pigment was proved to be indigo.The effects of temperature and rotational speed on indigo production were investigated,and 25 ℃ and 150 r·min-1 were selected as the most favorable conditions.Other variables during indigo production were optimized by response surface methodology as follows:inoculum 2.16(OD660),indole 200.55 mg·L-1 and pH 6.91.As a result,the yield of indigo obtained was up to 53.05 mg·L-1 under the optimal conditions,which was 179% higher than that of original conditions.
