Abstract:Objective To improve the diversity of secondary metabolites from actinomycetes, the solid-state fermentation technique was studied. Methods The culture medium including wheat bran, soybean meal, rice and corn grit, the initial moisture content and culturing time were optimized based on the diversity of secondary metabolites. Then effective secondary metabolite peaks from 35 soil actinomycetes under solid and liquid state fermentation were compared. Results The culture medium containing of wheat bran with 80% initial moisture, fermentation for 11 days were selected as the best fermentation conditions. The results showed that no effective secondary metabolite peaks were detected in the 5.7% and 37.1% of actinomycetes fermented with solid and liquid state manner, respectively; More effective secondary metabolite peaks were found in 82.9% of the strains with solid-state fermentation than that with liquid fermentation, 8.6% of the strains showed the same number of effective secondary metabolite peaks. Totally, the number of effective secondary metabolite peaks from 35 kinds of actinomycetes detected under solid-state fermentation was 3.5 times higher than that under liquid-state fermentation. Deducting the repeated metabolites in different strains, 34 kinds of metabolites were detected from 35 kinds of actinomycetes under solid-state fermentation, which were 2 times higher than that under liquid state fermentation. Conclusion Solid-state fermentation, which can improve the diversity of secondary metabolites, is expected to be a effective method to explore potential of medicinal actinomycetes in new secondary metabolites discovery.
