Abstract:Potassium clavulanate is one of the most widely used β-lactamase inhibitors in clinical application. A one-step crystallization process of potassium clavulanate was developed using potassium isooctanoate and clavulanic acid. Effects of factors, e.g. stirring rate and temperature on the quality and yield of potassium clavulanate were investigated. The optimized conditions are: potassium isooctanoate concentration = 0.56 molL-1, molar ratio of clavulanic acid to potassium isooctanoate = 1:1.2, addition rate of potassium isooctanoate solution = 4 mLmin-1. The stirring rate is 280 rmin-1 and temperature is 15 during the reaction process, and during the crystal growth process, the stirring rate is 180 rmin-1 and the cooling program is: 0.5min-1 for temperature from 15 to 10 and the nmaintained for 30 min. The system is kept cooling from 10 to 5 at 0.5min-1 and then maintained for 60 min at 5. The purity and yield of potassium clavulanate is over 99% and 73%, respectively. The impurity contents can meet the requirement of Chinese Pharmacopoeia 2015. Compared with two-step crystallization, this one-step crystallization process has similar yield but reduces operating steps and production costs. © 2018, Editorial Board of 'Journal of Chemical Engineering of Chinese Universities'.
