喷雾法制备载牛血清白蛋白海藻酸钙微球系统研究

Abstract:The alginate microspheres containing bovine serum albumin (BSA), chosen as a model protein, was prepared by spraying. The conditions of preparation were optimized by comparing the mean diameter of particles and the encapsulation efficiency of BSA. The results indicate that the encapsulation efficiency of BSA and the BSA loading are high (60% and 8%, respectively). The process of encapsulation has no effect on relative molecular mass, the molecular structure and the antigenicity of BSA by SDS-polyacrylamide gel electrophoresis (SDS-PAGE), Western-blot and circular dichroism (CD) spectrum. The in vitro release profile shows a controlled release of encapsulated BSA owing to the excellent pH responsive properties of alginate microspheres. Particularly in simulated gastric fluid, the release ratio and release amount of encapsulated BSA are low. Therefore, alginate microspheres are useful in intestinal-specific protein drugs delivery by protecting them from the destruction of gastric environment.
